目的:建立保健食品中菜油甾醇、豆甾醇和β-谷甾醇的含量测定方法。方法:采用气相色谱程序升温测定,以安捷伦DB-1701毛细管色谱柱(0.25μm×0.32 mm×30 m),汽化室温度280℃,检测器温度300℃,程序升温(起始温度100℃,保持2 min,以20℃·min-1升至270℃,保持15 min)。结果:菜油甾醇、豆甾醇和β-谷甾醇在该色谱条件下分离良好,其线性范围分别为51.08~255.4,20.86~104.3,16.13~80.64 mg·L-1,相关系数均>0.999 4,检出限分别为0.09%,0.08%,0.11%;样品加样回收率为88.5%~98.4%,RSD在0.23%~3.27%。结论:该方法灵敏、准确、可靠,能较好地分离上述3种植物甾醇,具有分析时间短、抗干扰性好、定量准确等优点,可为保健食品的质量标准提高提供科学数据。
Objective: To establish a method for the determination of campesterol, stigmasterol and β-sitosterol in health foods. Methods: The temperature of the sample was measured by GC-MS with the Agilent DB-1701 capillary column (0.25 μm × 0.32 mm × 30 m), the chamber temperature of 280 ℃, the detector temperature of 300 ℃, programmed temperature 2 min, raised to 270 ℃ at 20 ℃ · min-1 for 15 min). Results: The spectrums of campesterol, stigmasterol and β-sitosterol were separated well under the chromatographic conditions. The linear range was 51.08 ~ 255.4, 20.86 ~ 104.3, 16.13 ~ 80.64 mg · L-1, respectively. The limits of detection were 0.09%, 0.08% and 0.11%, respectively. The recoveries of samples were 88.5% -98.4% with RSDs of 0.23% -3.27%. Conclusion: The method is sensitive, accurate and reliable. It can separate the three phytosterols well and has the advantages of short analysis time, good anti-interference ability and accurate quantification. It can provide scientific data for the improvement of quality standard of health food.
