目的 :体外分离和诱导分化人胎肝干细胞。方法 :原代分离培养人胎肝干细胞集落 ,免疫细胞化学鉴定细胞集落分子标志物的表达 ;在体外因子作用下诱导干细胞定向分化为成熟肝脏细胞 ,对其生物学特性进行初步鉴定。结果 :从人胎肝组织中成功分离表达AFP、Albumin、Cytokeratin等标志物的胎肝干细胞集落 ;在体外因子作用下肝干细胞定向分化为成熟肝脏细胞。结论 :人胎肝中同样存在具有干细胞特性的原始细胞 ,体外可定向分化为成熟肝细胞。人肝干细胞的分离培养对于肝脏发育及其病理学的深入研究奠定了良好的基础。
OBJECTIVE: To isolate and induce human fetal liver stem cells in vitro. METHODS: Human fetal liver stem cell colonies were isolated and cultured in primary culture. Immunocytochemistry was used to identify the expression of cell colonies. The stem cells were induced to differentiate into mature liver cells under the action of exogenous factors. The biological characteristics of the cells were preliminarily identified. Results: Fetal liver stem cell colonies expressing AFP, Albumin, Cytokeratin and other markers were successfully isolated from human fetal liver tissue. Liver fibroblasts were differentiated into mature liver cells under the action of in vitro factors. Conclusion: There are also primitive stem cells in human fetal liver, which can be differentiated into mature hepatocytes in vitro. The isolation and culture of human hepatic stem cells has laid a good foundation for the further study of liver development and pathology.
