目的建立丙型肝炎病毒基因分型的基因芯片检测法,了解丙型肝炎病毒基因型与病毒复制及肝功能损害的关系。方法收集49例丙型肝炎患者血清标本,建立实验室基因芯片法检测丙型肝炎病毒基因型,采用荧光定量聚合酶链反应检测血清丙型肝炎病毒核糖核酸(HCVRNA)水平,同时检测丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)等肝功能相关指标。结果患者49例分为两组,1型丙型肝炎病毒有43例(87.76%),非1型丙型肝炎病毒有6例(12.24%)。1型与非1型相比较,两组患者血清HCVRNA、ALT水平没有明显差异(P>0.05),但血清AST水平差异有显著性(P<0.05)。结论基因芯片法可以对丙型肝炎病毒进行分型,具有临床应用价值。
Objective To establish a gene chip assay for genotyping hepatitis C virus to understand the relationship between hepatitis C virus genotypes and viral replication and impaired liver function. Methods The serum samples from 49 patients with hepatitis C were collected and the genotypes of hepatitis C virus were detected by the gene chip method in laboratory. The levels of serum hepatitis C virus RNA (HCVRNA) were detected by fluorescence quantitative polymerase chain reaction (PCR) Aminotransferase (ALT), aspartate aminotransferase (AST) and other indicators of liver function. Results The patients were divided into two groups: 43 cases (87.76%) of hepatitis C virus type 1 and 6 (12.24%) of non-type 1 hepatitis C virus. Compared with non-type 1, there was no significant difference in serum HCV RNA and ALT between the two groups (P> 0.05), but there was a significant difference in serum AST (P <0.05). Conclusion The gene chip method can classify the hepatitis C virus and has clinical value.
