为减损排斥反应性淋巴细胞克隆，采用异型双功能试剂ＳＰＤＰ、２ＩＴ连接ａｎｔｉＣＤ４、ＣＤ８单克隆抗体与天花粉毒素（ＴＣＳ），应用ＳｅｐｈａｃｒｙｌＳ２００分离游离ＴＣＳ，采用ＳＤＳＰＡＧＥ电泳及细胞毒性分析测定合成的免疫毒素的纯度和生物学活性。电泳结果显示，连接混合物中含有连接的免疫毒素、游离的单克隆抗体和ＴＣＳ，ＳｅｐｈａｃｒｙｌＳ２００可除去游离的ＴＣＳ。体外，免疫毒素可特异性杀伤大鼠脾细胞，其作用具有剂量依赖性。由此提示抗ＣＤ４、ＣＤ８免疫毒素可以诱导免疫无反应性
