高尔基体作为主要的分泌细胞器,其功能复杂。从细胞内分离出纯度较高的高尔基体有利于对其功能的深入研究。通过胃癌细胞SGC7901的大量体外培养,差速离心结合蔗糖密度梯度离心法纯化分离出高尔基体后,电镜及中性红超活染色进行形态学鉴定,高尔基体标志酶半乳糖基转移酶Galse及硫胺素焦磷酸酶TPPase活性测定进行纯度分析,首次从胃癌细胞SGC7901内成功分离出纯度较高的高尔基体,为对高尔基体功能的深入研究奠定基础。 Golgi as a major secretory organelle, its function is complex. The isolation of highly purified Golgi from the cell facilitates further study of its function. The SGC7901 cells were cultured in vitro and purified by differential centrifugation and sucrose density gradient centrifugation. Electron microscopy and neutral red supernuclear staining were used to identify the morphology of the Golgi apparatus. The Golgi marker of galactosyltransferase Galse and sulfur Purity analysis of TPPase activity assay for the first time successfully isolated from the SGC7901 gastric cancer cells with high purity of the Golgi apparatus for the in-depth study of the function of Golgi laid the foundation.
