本论文对滇型水稻品种玉优一号、HT - 7的愈伤组织诱导、基因转化及植株再生进行了研究。通过实验 ,将质粒 pCAMBI130 0上带有的PinⅡ抗虫基因导入水稻愈伤组织细胞中 ,获得了一批Hyg抗性植株 ,185株抗性苗经过PCR检测和叶片对潮霉素的抗性试验 ,证明其中有 5 9株为转基因阳性植株。结果显示 ,受体、农杆菌状态以及共培养方式等是影响基因转化效率的关键因素 ;以叶片对潮霉素的抗性作为转基因植株的快速筛选是可行的 ,大大提高了检测效率。
In this paper, callus induction, gene transformation and plant regeneration of Dian type rice variety Yuyou No.1, HT - 7 were studied. Through experiments, we obtained a batch of Hyg-resistant plants by introducing the PinⅡ-resistant gene with plasmid pCAMBI130 0 into rice callus cells. 185 resistant seedlings were tested by PCR and tested for resistance to hygromycin , Of which 59 were proved to be transgenic positive plants. The results showed that the status of receptor, Agrobacterium and co-culture were the key factors affecting the efficiency of gene transformation. It was feasible to use the resistance to hygromycin as the rapid screening of transgenic plants, which greatly enhanced the detection efficiency.
