目的 以骨形态发生蛋白 1 2 (BMP1 2 )基因转染骨髓间充质干细胞 ,探讨骨髓间充质干细胞定向分化为肌腱细胞的可行性。方法 将携带有BMP1 2基因的质粒pEGFP C1 ,采用电穿法转染骨髓间充质干细胞 ,转染后于荧光显微镜下观察绿色荧光蛋白 (GFP)表达情况。结果 转染后 6h可见细胞内有GFP表达 ,1 2h后达高峰 ,持续约 3天 ,其后有些细胞荧光开始减退。结论 BMP1 2基因转染骨髓间充质干细胞后 ,骨髓间充质干细胞内有GFP表达 ,提示细胞转染成功 ,骨髓向间充质干细胞定向分化为肌腱细胞具有可行性
OBJECTIVE: To investigate the feasibility of bone marrow mesenchymal stem cells (BMSCs) differentiating into tendon cells by transfecting bone marrow-derived mesenchymal stem cells with bone morphogenetic protein-12 (BMP1 2) gene. Methods Plasmid pEGFP C1 carrying BMP1 2 gene was transfected into bone marrow mesenchymal stem cells by electroporation. The expression of green fluorescent protein (GFP) was observed under a fluorescence microscope after transfection. Results The expression of GFP was observed within 6 hours after transfection and peaked at 12 hours after lasted for about 3 days. Afterwards, the fluorescence of some cells began to diminish. Conclusion BMP-2 gene transfection of bone marrow mesenchymal stem cells, bone marrow-derived mesenchymal stem cells GFP expression, suggesting that the successful transfection of cells, bone marrow mesenchymal stem cells tend to differentiate into tendon cells is feasible
