目的:建立鸭脚木皮中总三萜的含量测定方法并优选其提取工艺。方法:采用香草醛-高氯酸-浓硫酸法测定总三萜含量,检测波长540 nm。以总三萜提取率为指标,通过正交试验考察料液比、提取时间和提取次数对鸭脚木皮中总三萜提取工艺的影响。结果:以齐墩果酸为对照品,线性范围0.083 2~0.280 02 mg,加样回收率99.36%。最佳提取工艺为加10倍量水煎煮提取3次,每次1 h;总三萜平均提取率2.94%,RSD 0.5%。结论:建立的总三萜含量测定方法稳定可靠,优选的提取工艺合理可行,为鸭脚木皮有效部位的开发提供参考。
Objective: To establish a method for determination of total triterpenoids in duck foot veneer and optimize its extraction process. Methods: Total triterpene content was determined by vanillin - perchloric acid - concentrated sulfuric acid method. The detection wavelength was 540 nm. Taking the total triterpenoid extraction rate as an index, the orthogonal experiment was conducted to study the effects of the ratio of solid to liquid, the extraction time and the number of extraction on the total triterpenoid extraction process. Results: Oleanolic acid was used as a reference substance with a linear range of 0.083 2-0.280 02 mg and a recovery of 99.36%. The best extraction process was adding 10 times the amount of boiling water extraction 3 times, each time 1 h; total triterpene extraction rate 2.94%, RSD 0.5%. Conclusion: The established method for determination of total triterpenoids is stable and reliable, and the optimal extraction process is reasonable and feasible.
