目的探讨Cr(Ⅵ)染毒对A549细胞中hMSH2和p53mRNA表达的影响。方法体外培养A549细胞,实验组用脱氢抗坏血酸孵育90min,对照组常规培养,然后两组均用浓度分别为0.00、1.25、2.50、5.00μmol/L的Cr(Ⅵ)染毒。用实时聚合酶链反应(RT-PCR)检测A549细胞中hMSH2和p53mRNA的表达量。结果实验组与hMSH2和p53mRNA的相对表达量均低于对照组,差异均有统计学意义(P<0.05);Cr(Ⅵ)剂量依赖性地降低细胞中hMSH2和p53mRNA的表达;脱氢抗坏血酸预处理可以进一步降低暴露于Cr(Ⅵ)的A549细胞中hMSH2mRNA的表达。结论脱氢抗坏血酸在Cr(Ⅵ)致细胞毒性中起促进作用;Cr(Ⅵ)剂量依赖性地降低细胞中hMSH2和p53mRNA的表达,因此Cr(Ⅵ)可能参与肺癌细胞癌变过程。
Objective To investigate the effect of Cr (Ⅵ) on the expression of hMSH2 and p53 mRNA in A549 cells. Methods A549 cells were cultured in vitro. The experimental group was incubated with dehydroascorbic acid for 90 min. The control group was cultured routinely. Then the two groups were exposed to Cr (Ⅵ) at concentrations of 0.00, 1.25, 2.50 and 5.00 μmol / L respectively. The expression of hMSH2 and p53 mRNA in A549 cells was detected by real-time polymerase chain reaction (RT-PCR). Results The relative expression levels of hMSH2 and p53 mRNA in the experimental group were significantly lower than those in the control group (P <0.05). Cr (Ⅵ) decreased the expression of hMSH2 and p53 mRNA in a dose-dependent manner. Dehydroascorbate Treatment can further reduce hMSH2 mRNA expression in A549 cells exposed to Cr (VI). Conclusion Cr (Ⅵ) may play a role in carcinogenesis induced by Cr (Ⅵ). Cr (Ⅵ) may reduce the expression of hMSH2 and p53 mRNA in a dose - dependent manner.
