目的用LC-MS/MS法测定大鼠血浆中利福布丁的浓度。方法以替米沙坦为内标,加入0.2 mL血浆,用1 mL乙酸乙酯萃取,高速离心后上清液氮气吹干并用流动相复溶,取20μL进样检测。结果利福布丁的最低检测限为0.5 ng.mL-1,在0.3~9.6μg.mL-1内线性良好,低、中、高浓度的提取回收率均大于85%,批内、批间RSD低于10%,成功检测了大鼠血浆中利福布丁的血药浓度。结论所用方法灵敏度好、准确度高、分析时间短、样品处理更简便,适用于测定利福布丁的血药浓度。
Objective To determine the concentration of rifabutin in rat plasma by LC-MS / MS. Methods Telmisartan was used as an internal standard. 0.2 mL of plasma was added and extracted with 1 mL of ethyl acetate. After supernatant was centrifuged at high speed, the supernatant was air-dried with nitrogen and reconstituted with mobile phase. The results showed that the detection limit of rifabutin was 0.5 ng.mL-1, the linearity was good in 0.3 ~ 9.6 μg.mL-1, the recoveries were both higher than 85%. Intra-assay and inter-assay RSD Below 10%, the plasma concentration of rifabutin in rat plasma was successfully tested. Conclusion The method used has good sensitivity, high accuracy, short analysis time and simple sample processing. It is suitable for the determination of blood concentration of rifabutin.
