用带Ti质粒双载体 PGA492的生根农杆菌来转化热带豆科牧草矮笔花豆的叶和茎外植体。可获得86%的抗卡那霉素根系及23%的再生植株。这些转基因植株可育。生长正常或矮化,但也产生一些微小的形态变异。从首次再生植株的后代获得了正常表现型的转基因植株,证实再生植株具有活跃的新霉素磷酸转化酶活性和双载体 DNA 友 TL-DNA,并获得双载体和 TL-DNA 单独转移的证据。高频率培育出矮笔花豆转基因植株,大大改进了原先用致瘤农杆菌去臂系法所用的各种方法。
Agrobacterium rhizogenes with the Ti plasmid binary vector PGA492 was used to transform leaf and stem explants of the temperate forages of temperate leguminous plants. 86% of kanamycin resistant roots and 23% of regenerated plants were obtained. These transgenic plants are fertile. Normal or dwarf growth, but also produce some minor morphological changes. The transgenic plants with normal phenotype were obtained from the progeny of the first regenerated plants and confirmed that the regenerated plants had active neomycin phosphatase activity and double-carrier DNA-friend TL-DNA and obtained evidence of the dual carrier and TL-DNA alone. The high frequency of cultivating the short stylogenic transgenic plants has greatly improved the various methods previously used to detoxify the Agrobacterium tumefaciens.
