目的探讨WT1和eIF4E基因在儿童急性B淋巴细胞白血病中表达情况及意义。方法 73例儿童急性淋巴细胞白血病患者为白血病组,其中初诊患儿42例,完全缓解患儿23例,复发患儿8例。选择同期体检健康儿童20例为对照组,2组采用实时定量RT-PCR法检测WT1和eIF4E基因表达水平,并进行比较。结果白血病组WT1和eIF4EmRNA表达水平[208.9(48.9,3 902.5)、49.0(24.0,493.2)]明显高于对照组[33.7(14.0,99.7)、10.8(3.0,39.4)](P<0.05);初诊和复发患儿WT1基因表达水平[195.8(104.3,988.9)、1 649.9(687.4,4 502.5)]明显高于完全缓解患儿[90.5(44.9,317.0)](P<0.05),复发患儿明显高于初诊患儿(P<0.01);初诊患儿eIF4E基因表达水平[137.8(33.8,673.2)]明显高于完全缓解患儿[27.5(19.0,151.6)]和复发患儿[51.9(24.2,373.2)](P<0.01)。结论WT1基因参与了儿童急性B淋巴细胞白血病的初次发病和复发,eIF4E基因可能仅与初次发病有关。
Objective To investigate the expression and significance of WT1 and eIF4E in children with acute B lymphoblastic leukemia. Methods 73 cases of childhood acute lymphoblastic leukemia patients were leukemia patients, of which 42 cases of newly diagnosed children, 23 cases of complete remission in children, 8 cases of recurrent children. Twenty healthy children in the same period were selected as the control group. The expression of WT1 and eIF4E gene in the two groups were detected by real-time quantitative RT-PCR and compared. Results The expression levels of WT1 and eIF4E mRNA in leukemia group were significantly higher than those in the control group [208.9 (48.9,390.5), 49.0 (24.0, 493.2]] [33.7 (14.0,99.7), 10.8 (3.0, 39.4]]; The level of WT1 gene expression in newly diagnosed and relapsed children [195.8 (104.3,988.9), 1 649.9 (687.4,4 502.5)] was significantly higher than that in complete remission children [90.5 (44.9,317.0)] (P <0.05) The level of eIF4E gene expression in newly diagnosed children [137.8 (33.8,673.2)] was significantly higher than that in complete remission children [27.5 (19.0,151.6)] and relapsed children [51.9 (24.2 , 373.2)] (P <0.01). Conclusion The WT1 gene is involved in the initial onset and relapse of childhood acute B lymphoblastic leukemia. The eIF4E gene may be related to the first onset.
