目的:应用组织工程学原理,采用温度固化型合成水凝胶作为可注射用软骨的细胞载体,观察软骨的形成情况。方法:从兔耳中分离获取软骨细胞,将软骨细胞与水凝胶溶液混合,混合液的软骨细胞的浓度为50×106/ m l,注射于裸鼠背部皮下组织中,在室温下合成水凝胶由液态变为固态,包被软骨细胞于水凝胶体中。注射后4、8 周取材,通过大体标本观察、组织学观察软骨的形成情况。结果:注射后4 周在皮下形成稍硬的结节,8 周时形成的结节质地坚韧,切面色泽亮白。组织学观察,4 周标本中即有软骨形成,但不成熟,8 周标本中有成熟软骨形成,番红 O染色呈强阳性。结论:温度固化型合成水凝胶作为软骨细胞载体,可以通过注射的方法建造可注射性软骨,具有广阔的应用前景。
OBJECTIVE: To observe the formation of cartilage by using the principle of tissue engineering, using temperature-curing synthetic hydrogel as a cell carrier of injectable cartilage. Methods: Chondrocytes were isolated from rabbit ears and chondrocytes were mixed with hydrogel solution. The concentration of chondrocytes in the mixture was 50 × 106 / ml. The chondrocytes were injected into the subcutaneous tissue of the back of nude mice and synthesized water at room temperature The gel changes from a liquid state to a solid state, coating the chondrocytes in the hydrogel. At 4 and 8 weeks after injection, the specimens were observed and the formation of cartilage was observed by histological observation. Results: Slightly harder nodules were formed 4 weeks after injection. The nodules formed at 8 weeks were tough and the color of the section was bright. Histological observation showed that cartilage was formed in 4-week specimens, but immature. There was mature cartilage in 8-week specimens and Safranine-0 staining was strongly positive. Conclusion: As a chondrocyte carrier, temperature-curable synthetic hydrogels can be injected into cartilage and have broad application prospects.
