目的:探讨胃癌中医病理因素特征与胃癌组织抑癌基因ING1甲基化及CD133、CD44基因表达间的关系。方法:采集100例胃癌术前临床资料,进行中医病理因素的提取,探求其病理因素特征,并采用甲基化特异性PCR(MS-PCR)测其手术切除的胃癌组织和相应的癌旁组织中ING1甲基化水平,RT-PCR和免疫组织化学法测CD133、CD44水平,探讨ING1甲基化对CD133、CD44表达水平的影响,并采用二元logistic回归分析胃癌病理因素与ING1甲基化状况、CD133和CD44基因表达水平的相关性。结果:胃癌病理因素主要为气虚、痰、瘀、气滞;胃癌组织和癌旁组织ING1甲基化和CD133、CD44表达差异有显著性意义(P<0.05);胃癌组织ING1甲基化状况对CD133、CD44表达产生显著影响(P<0.05);ING1甲基化状况、CD133和CD44表达水平对胃癌中医病理因素气虚、痰、瘀产生显著影响(P<0.05)。结论:胃癌病理因素气虚、痰、瘀与ING1甲基化状况、CD133和CD44表达水平具有显著相关性。
Objective: To investigate the relationship between the pathological features of gastric cancer and the methylation of tumor suppressor gene ING1 and the expression of CD133 and CD44 in gastric cancer. Methods: Preoperative clinical data of 100 cases of gastric cancer were collected. The pathological factors of traditional Chinese medicine were extracted and their pathological factors were explored. Methylation-specific PCR (MS-PCR) was used to detect the resected gastric cancer tissues and corresponding paracancerous tissues ING1 methylation levels, RT-PCR and immunohistochemistry were used to measure the levels of CD133 and CD44, and to explore the effect of ING1 methylation on the expression of CD133 and CD44. Binary logistic regression was used to analyze the correlation between the pathological factors of gastric cancer and ING1 methylation Status, CD133 and CD44 gene expression levels of the correlation. Results: The main pathological factors of gastric cancer were Qi deficiency, phlegm, blood stasis and qi stagnation. The methylation of ING1 and the expressions of CD133 and CD44 in gastric cancer tissue and paracancerous tissue were significantly different (P <0.05). The methylation status of ING1 in gastric cancer CD133 and CD44 (P <0.05). The methylation status of ING1, the expression of CD133 and CD44 were significantly affected in the pathogenesis of qi deficiency, phlegm and stasis syndrome of gastric cancer (P <0.05). Conclusion: There is a significant correlation between the pathological factors of Qi deficiency, phlegm and blood stasis and the status of ING1 methylation, the expression of CD133 and CD44 in gastric cancer.
