研究凋亡调控基因 bcl- 2和 bax m RNA在急性白血病 ( AL)中的表达、相互关系和临床意义 ,应用半定量逆转录 -聚合酶链反应 ( RT- PCR)和 Southern blot技术检测 3 2例 AL患儿 bcl- 2和 bax m RNA水平及其比值。结果显示 ,AL患儿骨髓细胞中 bcl- 2 m RNA的表达明显高于对照组 ( P<0 .0 1) ,bax m RNA的表达明显降低( P<0 .0 1)。治疗后 ,bcl- 2 m RNA在两组间的表达无差异 ( P>0 .0 5 )。分析 bcl- 2 /baxm RNA表达比值显示 ,高比值组 NR率显著高于低比值组 ( P<0 .0 1)。结论 :AL的发病与凋亡调控基因 bcl- 2和 bax的异常表达有关。bcl- 2和 bax m RNA及其比值可作为判断 AL预后及指导治疗的主要监测指标
To investigate the expression, relationship and clinical significance of apoptosis-regulatory genes bcl-2 and bax m RNA in acute leukemia (AL). Semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Southern blot techniques were used to detect 3 2 . Levels of bcl-2 and bax m RNA in children with AL and their ratios. The results showed that the expression of bcl-2 mRNA in bone marrow cells of AL children was significantly higher than that in the control group (P<0.01), and the expression of bax mRNA was significantly decreased (P<0.01). After treatment, there was no difference in bcl-2 mRNA expression between the two groups (P>0.05). Analysis of the bcl-2/baxm RNA expression ratio showed that the NR rate was significantly higher in the high ratio group than in the low ratio group (P < 0.01). Conclusion : The pathogenesis of AL is related to the abnormal expression of apoptosis regulating genes bcl-2 and bax. Bcl-2 and bax m RNA and their ratios can be used as the main monitoring indicators to predict the prognosis and treatment of AL
