目的:建立白花蛇舌草药材的HPLC指纹图谱。方法:采用高效液相色谱-二极管阵列检测法,选用Diamonsil C18(250 mm×4.6 mm,4μm)色谱柱,Easy GuardⅡC18为保护柱,甲醇-0.1%冰醋酸溶液为流动相梯度洗脱,流速1.0 ml·min-1,检测波长254 nm,柱温40℃。采用中国药典委员会颁布的《中药色谱指纹图谱相似度评价系统(2012.130723)》评价不同产地白花蛇舌草图谱的相似性。结果:采用10批白花蛇舌草药材建立HPLC指纹图谱,方法学符合指纹图谱技术要求,10批白花蛇舌草的相似度在0.766~0.985之间。结论:该指纹图谱方法稳定、准确、可靠,为白花蛇舌草药材质量标准的制定提供了科学依据。
Objective: To establish HPLC fingerprints of Hedyotis diffusa. Methods: Diamonsil C18 (250 mm × 4.6 mm, 4 μm) column was used as the guard column and methanol-0.1% glacial acetic acid was used as the mobile phase gradient elution at a flow rate of 1.0 ml · min-1, detection wavelength 254 nm, column temperature 40 ℃. The similarity of chromatograms of Hedyotis diffusa in different areas was evaluated using the “similarity evaluation system of chromatographic fingerprints of traditional Chinese medicines (2012.130723)” promulgated by the Chinese Pharmacopoeia Commission. Results: The HPLC fingerprints of 10 Hedyotis diffusa herbs were established. The method met the requirements of fingerprinting technique. The similarity of 10 batches of Hedyotis diffusa was between 0.766 and 0.985. Conclusion: The fingerprinting method is stable, accurate and reliable and provides a scientific basis for the formulation of the quality standard of Hedyotis diffusa.
