目的探讨靶向PEG10的siRNA真核表达载体(psiRNA-PEG10)对裸鼠人肝癌移植瘤生长的影响。方法建立人肝癌细胞HepG2的荷瘤裸鼠模型,在接种后第14天分别将psiRNA空载体和psiR-NA-PEG10瘤内多点注射进行治疗,观察肿瘤大小变化、组织形态学及超微病理改变。结果psiRNA-PEG10治疗组肿瘤生长受到明显抑制,与生理盐水组和psiRNA组比较差异有显著性(P<0.001),抑瘤率明显高于其他两组,电镜下可见明显的细胞凋亡。结论靶向PEG10的siRNA真核表达载体对裸鼠肝癌移植瘤生长有抑制作用。
Objective To investigate the effect of psiRNA-PEG10 siRNA targeting PEG10 on the growth of human hepatocellular carcinoma in nude mice. Methods Human hepatoma HepG2 tumor-bearing nude mice model was established. On the 14th day after inoculation, psiRNA empty vector and psiR-NA-PEG10 were injected intratumorally for multiple injections. The changes of tumor size, histomorphology and ultrastructure were observed change. Results The tumor growth of psiRNA-PEG10 group was significantly inhibited. Compared with saline group and psiRNA group, the difference was significant (P <0.001), and the tumor inhibition rate was significantly higher than the other two groups. The apoptosis of cells was observed under electron microscope. CONCLUSION: siRNA targeting PEG10 can inhibit the growth of hepatocellular carcinoma in nude mice.
