目的:探讨贞芪酪蛋白复合肽对Bel-7402人肝癌细胞分化的诱导作用,及对其端粒酶活性的影响。方法:贞芪酪蛋白复合肽给正常大鼠灌胃,采血制备药物血清。以正常鼠血清为阴性对照,维甲酸(RA)为阳性对照,将药物血清温育Bel-7402人肝癌细胞。噻唑蓝(MTT)比色法测定细胞增殖,图像分析系统测定细胞核质比,放射免疫法测定甲胎蛋白(AFP)含量,动态比色法测定γ-谷氨酰转肽酶(-γGT)和碱性磷酸酶(ALP)活性,多聚酶链反应—酶联免疫吸附法(PCR-ELISA法)检测细胞端粒酶活性。结果:贞芪酪蛋白复合肽药物血清可抑制人肝癌细胞的增殖,显著降低细胞核质比,抑制细胞分泌AFP,降低细胞-γGT活性,升高ALP活性,并有显著抑制人肝癌细胞端粒酶活性的作用。结论:贞芪酪蛋白复合肽具有诱导人肝癌细胞分化的作用,其主要机理可能是抑制了Bel-7402细胞端粒酶活性。
OBJECTIVE: To investigate the induction of differentiation of Bel-7402 human hepatocellular carcinoma cells and its effect on telomerase activity by the combination of sarcoside casein complex peptide. METHODS: The scutellarin casein complex peptide was given to normal rats by gavage, and blood was collected to prepare drug serum. Normal rat serum was used as a negative control. Retinoic acid (RA) was a positive control. The drug serum was incubated with Bel-7402 human hepatoma cells. MTT colorimetric method was used to measure cell proliferation. The image analysis system was used to determine the ratio of nucleus to cytoplasm. Radioimmunoassay was used to determine the alpha-fetoprotein (AFP) content. The dynamic colorimetric method was used to determine γ-glutamyl transpeptidase (-γGT). Alkaline phosphatase (ALP) activity, polymerase chain reaction-enzyme-linked immunosorbent assay (PCR-ELISA) was used to detect cell telomerase activity. Results: The serum of casein combined peptide could inhibit the proliferation of human hepatoma cells, significantly reduce the ratio of cytoplasm to cytoplasm, inhibit the secretion of AFP, decrease the activity of cell-γGT, increase the activity of ALP, and significantly inhibit the telomerase activity of human hepatoma cells. The role of activity. CONCLUSION: Casein complex peptide can induce differentiation of human hepatoma cells. The main mechanism may be inhibition of telomerase activity in Bel-7402 cells.
