[Objective] To establish a method for the extraction and determination of polysaccharides from FRUCTUS CHAENOMELIS. [Methods] Ethanol reflux method was adopted to extract polysaccharides from FRUCTUS CHAENOMELIS. Then, the content of polysaccharides was determined by the method of C6H5OH phenol-H2SO4. [Results] The linear regression equation was Y=0.008 8X-0.010 9, R2=0.992 3. There was good linearity between concentration and absorbance of polysaccharides in the range of 1.0 to 5.0 μg/ml. Through twice extraction, the content of polysaccharides was 13.21% in defatted sample and 9.26% in original sample. The average recovery was 97.04% with RSD of 3.52%. [Conclusion] This method is simple, stable, precise and reproducible, which is suitable for extraction and determination of polysaccharides from FRUCTUS CHAENOMELIS.
