目的:探讨补肾温阳祛风散寒法对肾阳虚型围绝经期模型大鼠ER、VEGF、HSP70的影响及作用机制。方法:选用自然老化的围绝经期雌性SD大鼠,肌注氢化可的松构建肾阳虚型围绝经期大鼠模型,将模型大鼠随机分为更年汤组、教材方组、替勃龙组、生理盐水组;另选青年组、老年组SD雌性大鼠作为对照组。各组给予相应药物治疗15 d,于第16天处死大鼠,分离摘取双侧子宫,称重了解子宫湿重及子宫指数,采用病理切片观察大鼠子宫形态学变化,免疫组化及原位杂交检测方法观察大鼠子宫雌激素受体(ER)、血管内皮生长因子(VEGF)、热休克蛋白70(HSP70)及信使mRNA(ERmRNA、VEGFmRNA、HSP70mRNA)的阳性表达情况。结果:1更年汤组、教材方组、替勃龙组能明显增加子宫湿重、子宫指数,有统计学意义(P<0.05),更年汤组与替勃龙组、教材方组比较差异有统计学意义(P<0.05);2更年汤组、教材方组、替勃龙组能抑制子宫萎缩,有显著性差异(P<0.01),更年汤组与替勃龙组比较无统计学意义(P>0.05);3更年汤组、教材方组、替勃龙组能增加ER、ERmRNA、VEGF、VEGFmRNA阳性表达,有统计学意义(P<0.05),但教材方组的上调低于更年汤组及替勃龙组(P<0.05),替勃龙组与更年汤组之间比较无显著差异(P>0.05);4更年汤组、替勃龙组能增加HSP70及HSP70mRNA阳性表达,有统计学意义(P<0.05),更年汤组与替勃龙组比较无显著性差异(P>0.05)。结论:自拟更年汤可能通过上调ER、VEGF、HSP70蛋白及基因在子宫中的阳性表达而发挥雌激素样作用,从而达到改善病理状态下的子宫的形态结构及功能的目的。
Objective: To investigate the effect and mechanism of BushenYangyang Qufeng Sanhan method on ER, VEGF, HSP70 in rats with perimenopausal kidney deficiency syndrome. Methods: A spontaneous perimenopausal female Sprague-Dawley (SD) rats were randomly divided into three groups: control group, treatment group, Dragon group, saline group; another young group, aged SD female rats as a control group. The rats in each group were given the corresponding drugs for 15 days. Rats were sacrificed on the 16th day. The bilateral uterus were isolated and weighed and the uterine wet weight and uterine index were weighed. The morphological changes of the uterus were observed by immunohistochemistry and immunohistochemistry The hybridization test was used to observe the positive expression of estrogen receptor (ER), vascular endothelial growth factor (VEGF), heat shock protein 70 (HSP70) and messenger mRNA (ERmRNA, HSP70mRNA) in rat uterus. Results: 1 Gengnian decoction group, teaching materials group and tibolone group can significantly increase uterine wet weight and uterine index (P <0.05), compared with those of Gengnian decoction group and Tibolone group The difference was statistically significant (P <0.05); 2 Gengnian decoction group, teaching material group, tibolone group can inhibit uterine atrophy, there was significant difference (P <0.01) (P> 0.05) .3 The Gengnian decoction group, the teaching materials group and the tibolone group can increase the expression of ER, ERmRNA, VEGF and VEGFmRNA, with statistical significance (P <0.05) (P <0.05). There was no significant difference between tibolone group and Gengnian decoction group (P> 0.05); 4 Gengnian decoction group and Tibolone group (P <0.05). There was no significant difference between Gengnian decoction group and tibolone group (P> 0.05). CONCLUSION: Self-made Decoction can play an estrogen-like role by up-regulating the expression of ER, VEGF and HSP70 in uterus and thus improving the morphology and function of the uterus in pathological conditions.
