目的:优选白术内酯Ⅰ脂质体最佳制备工艺及处方。方法:采用薄膜分散法制备白术内酯Ⅰ脂质体,以包封率为评价指标,采用正交设计优化处方和制备工艺,采用HPLC法测定其包封率。结果:最佳处方工艺为:卵磷脂与胆固醇的质量比为6:1,卵磷脂与白术内酯Ⅰ质量比为20:1,PBS缓冲溶液pH为7.5。结论:经优化得到的白术内酯Ⅰ脂质体处方合理,工艺可行,包封率较高。
Objective: To optimize the preparation and formulation of liposomal Atractylenolide I. Methods: Atractylenolide I liposome was prepared by film dispersion method. The encapsulation efficiency was evaluated. The formulation and preparation process were optimized by orthogonal design. The encapsulation efficiency was determined by HPLC. RESULTS: The optimum formulation process was that the mass ratio of lecithin to cholesterol was 6:1, the mass ratio of lecithin to atractylenolide I was 20:1, and the pH of PBS buffer solution was 7.5. Conclusion: The optimized liposomes of atractylenolide I have a reasonable prescription, feasible process and high entrapment efficiency.
