目的研究阴道超声对绒毛滋养细胞凋亡的影响及随着时间的推移其凋亡指数的变化,同时探索Bcl-2、活化Caspase-3在滋养细胞凋亡中的表达。方法收集2012年10月至2013年8月深圳市南山区蛇口人民医院收治的行人工流产前接受阴道超声照射的患者100例,设为观察组,根据阴道超声照射后流产时间的不同随机分为观察组1、观察组2、观察组3、观察组4,每组各25例。经阴道超声持续照射5分钟分别于24h、3d、5d、7d后行人工流产收集绒毛标本,另选择同期收治的未经超声照射行人工流产的患者25例设对照组。采用TUNEL法观察经阴道超声辐照后,不同天数的人胚绒毛滋养细胞的凋亡情况及随着时间的推移其凋亡指数的变化规律。同时用免疫组化SABC法检测相应滋养细胞中凋亡相关蛋白Bcl-2及活化Caspase-3表达水平的变化。结果经阴道超声照射后随取胚天数的后延,凋亡指数呈下降趋势。Bcl-2蛋白表达量逐渐增加,活化Caspase-3蛋白表达量逐渐减少。照射后1、3、5d与对照组比较差异有统计学意义(P<0.05);但在照射后第7d与对照组比较差异无统计学意义(P>0.05)。结论Bcl-2蛋白和活化Caspase-3蛋白在细胞凋亡转导通路中发挥了重要的调控作用,同时证实了阴道超声辐射对绒毛滋养细胞凋亡有一定的影响,但呈近期可逆性改变。
Objective To study the effect of vaginal ultrasound on the apoptosis of villous trophoblast cells and the change of apoptosis index with the passage of time, and to explore the expression of Bcl-2 and activated Caspase-3 in trophoblast apoptosis. Methods 100 patients who underwent vaginal ultrasound before artificial abortion were treated in Shekou People’s Hospital of Nanshan District, Shenzhen from October 2012 to August 2013 were selected as the observation group and randomly divided according to the time of vaginal ultrasound irradiation Observation group 1, observation group 2, observation group 3, observation group 4, each group 25 cases. Vaginal ultrasound continuous irradiation for 5 minutes were 24h, 3d, 5d, 7d after artificial abortion to collect villus specimens, the other selected the same period of treatment without artificial abortion in patients with 25 cases of control group. TUNEL method was used to observe the changes of apoptotic index of human embryos villus trophoblast cells after transvaginal ultrasound irradiation for different days. At the same time, the expression of Bcl-2 and Caspase-3 in the corresponding trophoblasts was detected by immunohistochemical SABC method. Results After transvaginal ultrasound irradiation, the apoptotic index decreased with the number of embryos retrieved. The expression of Bcl-2 protein gradually increased, and the expression of activated Caspase-3 protein gradually decreased. There was significant difference between the control group and the 1st, 3rd, 5th day after irradiation (P <0.05), but there was no significant difference between the control group and the 7th day after irradiation (P> 0.05). Conclusions Bcl-2 protein and activated Caspase-3 protein play an important regulatory role in apoptosis pathway. At the same time, it is confirmed that vaginal ultrasound radiation has a certain influence on the apoptosis of villous trophoblast cells, but it is reversible.
