目的:观察和探讨灵芝孢子粉对癫痫大鼠脑组织细胞因子IL-6含量变化的影响。方法:10周龄Wistar雄性大鼠30只,体重(150±10)g,随机分成3组,每组10只,分别是灵芝孢子粉组、癫痫模型组和空白对照组。灵芝孢子粉组和癫痫模型组用戊四氮(PTZ)亚惊厥剂量(32mg/kg),每日1次,腹腔注射28d,停药1周,再用相同剂量的PTZ测试,凡显示连续5次2级以上惊厥的大鼠为达到点燃标准,空白对照组用同体积生理盐水腹腔注射。同时灵芝孢子粉组经灌胃给予灵芝孢子粉(灌胃前禁食6h),灌胃体积为1.0ml/次;癫痫模型组和空白对照组经灌胃给予同等体积的生理盐水。实验结束处死动物,在低温条件下迅速取出鼠脑,用放射免疫法测定脑组织细胞因子IL-6含量变化。结果:灵芝孢子粉组与癫痫模型组动物相比脑组织IL-6明显降低(P<0.05)。结论:灵芝孢子粉能够有效降低癫痫大鼠脑组织细胞因子IL-6水平,纠正免疫失调而起到抗癫痫作用。
OBJECTIVE: To observe and investigate the effect of Ganoderma lucidum spore powder on the change of cytokine IL-6 in brain tissue of epileptic rats. METHODS: Thirty Wistar male rats aged 10 weeks were weighed (150 ± 10) g and randomly divided into 3 groups of 10 in each group: Ganoderma lucidum spore powder group, epilepsy model group and blank control group. The ganoderma lucidum spore powder group and epilepsy model group were treated with pentylenetetrazol (PTZ) subconvulsive dose (32 mg/kg) once a day, intraperitoneal injection for 28 days, withdrawal for 1 week, and then the same dose of PTZ test, where continuous 5 The second-level or more convulsive rats to achieve ignition standards, the blank control group was injected intraperitoneally with the same volume of saline. At the same time, Ganoderma lucidum spore powder was administered by gavage to the spore powder of Ganoderma lucidum (fed 6 h before gavage), and the volume of gavage was 1.0 ml/time. The epilepsy model group and the blank control group were given the same volume of normal saline by gavage. Animals were sacrificed at the end of the experiment and the brains of rats were quickly removed under low temperature conditions. Changes in cytokine IL-6 levels in brain tissue were determined by radioimmunoassay. RESULTS: Compared with epilepsy model animals, IL-6 in brain tissue of ganoderma lucidum spore powder group was significantly lower (P<0.05). Conclusion: Ganoderma lucidum spore powder can effectively reduce the level of cytokine IL-6 in the brain of epileptic rats, correct immune disorders and play an anti-epileptic effect.
