采用亲和层析法纯化的AFP抗原免疫的BALB/c鼠脾细胞与SP2/0小鼠骨髓瘤细胞系融合,获三株分泌抗人AFP单克隆抗体(McAb)的杂交瘤细胞系.用ELISA阻断试验和夹心ELISA试验作特异性鉴定,杂交瘤细胞株染色体的众数为104条,三株细胞系McAb均属小鼠IgG1型.经体外连续传代3个月及冻存一年复苏后均能持续分泌抗AFP抗体
The BALB / c mouse spleen cells immunized with the AFP antigen purified by affinity chromatography were fused with the SP2 / 0 mouse myeloma cell line and three hybridoma cell lines secreting anti-human AFP monoclonal antibody (McAb) were obtained. Specificity was identified by ELISA and sandwich ELISA. The number of chromosomes in the hybridoma cell line was 104, and all the three cell lines McAb were mouse IgG1.
