目的:探讨抵挡汤早期干预对2型糖尿病大鼠血管内皮细胞凋亡的影响。方法:将SD大鼠随机分为正常对照组,糖尿病模型组,抵挡汤早、中、晚期干预组,二甲双胍组,辛伐他汀组。酶联免疫吸附实验(ELISA)检测活性氧(ROS)含量,实时荧光定量PCR检测凋亡诱导因子(AIF)、凋亡蛋白激酶激活因子-1(Apaf-1)、半胱天冬肽酶-3(Caspase-3)基因表达水平。结果:与正常对照组比较,糖尿病模型组ROS含量,AIF、Apaf-1、Caspase-3基因表达水平均显著升高(P<0.05);与糖尿病模型组比较,抵挡汤早、中期干预组,二甲双胍组及辛伐他汀组ROS含量,AIF、Apaf-1、Caspase-3基因表达水平均显著下降(P<0.05),且在抵挡汤早期干预组中AIF、Apaf-1、Caspase-3基因表达水平显著低于中、晚期干预组(P<0.05)。结论:抵挡汤早期干预可以通过抗氧化应激减少血管内皮细胞凋亡。
Objective: To investigate the effect of Zhongdan Decoction on vascular endothelial cell apoptosis in type 2 diabetic rats. Methods: SD rats were randomly divided into normal control group, diabetic model group, resist early, middle and late intervention group, metformin group and simvastatin group. The level of reactive oxygen species (ROS) was detected by enzyme-linked immunosorbent assay (ELISA). Apoptosis-inducing factor (AIF), Apaf-1 and caspase- 3 (Caspase-3) gene expression levels. Results: Compared with the normal control group, the levels of ROS, AIF, Apaf-1 and Caspase-3 in diabetes model group were significantly increased (P <0.05); Compared with the diabetic model group, The levels of ROS, AIF, Apaf-1 and Caspase-3 in metformin and simvastatin groups were significantly decreased (P <0.05), and the expression of AIF, Apaf-1 and Caspase-3 in the early intervention group The level was significantly lower than the middle and late intervention group (P <0.05). Conclusion: The early resisting of soup can reduce the apoptosis of vascular endothelial cells through anti-oxidative stress.
