目的探讨Gal-1蛋白促胃癌细胞侵袭转移的可能机制。方法采用0、1和5μg/m L的Gal-1蛋白体外刺激胃癌细胞,利用肿瘤细胞侵袭模型分析Gal-1蛋白对胃癌细胞侵袭迁移能力的影响;利用WB实验和明胶酶谱法,分别检测Gal-1蛋白刺激前后胃癌细胞中MMP-9的表达及活性变化,分析Gal-1促胃癌转移的可能分子机制。结果细胞迁移实验显示,胃癌细胞BGC-823经1和5μg/m L的Gal-1蛋白刺激后穿膜细胞数分别为(117±8.19)和(167±7.55)个,明显高于0μg/m L组的(67±4.58)个(P<0.05);胃癌7 901细胞经1和5μg/m L的Gal-1蛋白刺激后穿膜细胞数分别为(151±5.13)和(190.3±6.8)个,高于0μg/m L组的(87±6.56)个(P<0.05)。细胞侵袭实验显示,BGC-823经1和5μg/m L的Gal-1蛋白刺激后穿膜细胞数分别为(51±3.6)和(76.7±9.07)个,明显高于0μg/m L组的(22.7±4.16)个(P<0.05);胃癌7 901细胞经1和5μg/m L的Gal-1蛋白刺激后穿膜细胞数分别为(74.0±7.21)和(105.3±11.37)个,高于0μg/m L组的(42.3±6.66)个(P<0.05)。WB实验显示,经Gal-1蛋白刺激后,胃癌细胞中MMP-9的表达增高。明胶酶谱实验显示,Gal-1蛋白刺激后,MMP-9酶活性增强。结论 Gal-1蛋白能促进胃癌细胞的侵袭迁移能力,可能与其能增强胃癌细胞中的MMP-9表达和活性有关。
Objective To investigate the possible mechanism of Gal-1 protein in invasion and metastasis of gastric cancer cells. Methods Gal-1 protein of 0, 1 and 5 μg / mL was used to stimulate gastric cancer cells in vitro. The effect of Gal-1 protein on invasion and migration of gastric cancer cells was analyzed by tumor cell invasion model. WB and gelatin zymography were used to detect the effect of Gal- The expression and activity of MMP-9 in gastric cancer cells before and after Gal-1 stimulation were analyzed. The possible molecular mechanism of Gal-1 in gastric cancer metastasis was analyzed. Results The results of cell migration showed that the numbers of transmembrane cells in gastric cancer cells BGC-823 were (117 ± 8.19) and (167 ± 7.55) after stimulation with 1 and 5 μg / mL Gal-1 protein, respectively, which were significantly higher than 0 μg / m (P <0.05). The numbers of transmembrane cells in gastric cancer 7901 cells treated with Gal-1 at 1 and 5 μg / mL were (151 ± 5.13) and (190.3 ± 6.8) (87 ± 6.56) higher than 0μg / m L group (P <0.05). Cell invasion assay showed that the number of transmembrane cells in BGC-823 cells treated with 1 and 5 μg / mL Gal-1 protein was (51 ± 3.6) and (76.7 ± 9.07), respectively, which was significantly higher than that in 0 μg / mL (22.7 ± 4.16), respectively (P <0.05). The numbers of transmembrane cells in 7901 cells treated with Gal-1 at 1 and 5 μg / mL were (74.0 ± 7.21) and (105.3 ± 11.37) (42.3 ± 6.66) in the 0 μg / m L group (P <0.05). WB experiments showed that the expression of MMP-9 in gastric cancer cells was increased after Gal-1 protein stimulation. Gelatin zymography experiments show that, Gal-1 protein stimulation, MMP-9 enzyme activity increased. Conclusions Gal-1 protein can promote the invasion and migration of gastric cancer cells, which may be related to the enhancement of MMP-9 expression and activity in gastric cancer cells.
