目的探讨肿节风总黄酮对体外培养巨核细胞增殖的影响及其效应动力学。方法肿节风总黄酮(0.39g·kg-1)单次灌胃给予正常SD大鼠,给药后不同时间点采血制备含药血浆。采用兔抗大鼠血小板相关抗体建立大鼠骨髓巨核细胞增殖障碍模型,并将不同时间点的含药血浆作用于该模型,以巨核细胞数、细胞上清液中血小板生成素(TPO)、转化生长因子-β1(TGF-β1)含量为效应指标,绘制时间-效应动力学曲线,计算相关动力学参数。结果肿节风总黄酮含药血浆可以显著提升巨核细胞数,肿节风总黄酮对巨核细胞数、上清液中TPO及TGF-β1的效应半衰期分别为:495.52,665.13和537.46 min,达峰时间分别为:60,90和40 min,平均驻留时间分别为:224.60,242.37和231.10 min,其中巨核细胞数与TPO含量的效应曲线之间有较好的相关性,时间上稍有滞后现象。结论肿节风总黄酮含药血浆可显著促进大鼠骨髓巨核细胞的增殖,可能与培养体系中TPO的浓度密切相关。
Objective To investigate the effects of total flavonoids of Typhoid fever on the proliferation of megakaryocytes in vitro and their effects on kinetics. Methods Effervescent total flavonoids (0.39g · kg-1) were intragastrically given to normal SD rats at different time points after administration of drug-containing plasma preparation. Rat anti-rat platelet-associated antibody was used to establish a model of proliferative disorders of rat bone marrow megakaryocytes, and the drug-containing plasma at different time points was applied to the model. The number of megakaryocytes, the concentration of TPO in the cell supernatant, Growth factor-β1 (TGF-β1) content as an effect index, draw the time-response kinetic curve, calculate the relevant kinetic parameters. Results The total flavonoids in swollen-air-fistulae could significantly increase the number of megakaryocytes. The half-life of TPO and TGF-β1 in the supernatant was 495.52, 665.13 and 537.46 min, respectively, The time was 60, 90 and 40 min respectively. The average residence time was 224.60, 242.37 and 231.10 min, respectively. There was a good correlation between the number of megakaryocytes and the effect curve of TPO content, with a slight lag in time . Conclusions The total flavonoids-containing plasma in the treatment group may promote the proliferation of the bone marrow megakaryocytes significantly, which may be closely related to the concentration of TPO in the culture system.
