目的探讨维吾尔族肥胖患者内脂素、脂联素(APN)在腹部网膜、皮下脂肪组织的表达和血清水平改变及其与体重指数(BMI)、腰臀围比(WHR)和血脂的关系。方法用半定量RT-PCR方法检测41例肥胖患者和20例非肥胖对照脂肪组织内脂素和APNmRNA表达水平,ELISA法检测血清内脂素和APN浓度,并测量腰围(WC)、臀围(HC)、血压(BP)、空腹血糖(FPG)和血脂等。结果①网膜脂肪组织内脂素mRNA表达水平在肥胖组与非肥胖对照组之间差异无显著性,肥胖组APNmRNA表达水平显著低于非肥胖对照组(P<0.05);内脂素、APN在两组网膜和皮下脂肪组织mRNA的表达水平均无显著性部位差异;血清内脂素肥胖组显著高于非肥胖对照组(P<0.05),APN在两组之间比较差异无显著性。②肥胖组舒张压(DBP)和收缩压(SBP)均显著高于非肥胖对照组(P<0.01和P<0.01),高密度脂蛋白(HDL-C)明显低于非肥胖对照组(P<0.05)。③网膜脂肪组织内脂素mRNA表达量与血清内脂素、BMI和TG显著正相关,APNmRNA表达量与血清APN显著正相关,与BMI、LDL-C显著负相关。结论维吾尔族肥胖患者血清内脂素水平升高,网膜和皮下脂肪组织内脂素mRNA表达水平无显著性部位差异;APNmRNA在网膜脂肪组织的表达显著降低,内脂素和脂联素可能在维吾尔人中心性肥胖相关的代谢紊乱性疾病中起一定作用。
Objective To investigate the expression of adiponectin and adiponectin (APN) in the abdominal omentum and subcutaneous adipose tissue and their serum levels in obese Uygur patients and their relationship with body mass index (BMI), waist-hip ratio (WHR) and serum lipids . Methods The expression of adiponectin and APN mRNA in 41 cases of obese patients and 20 cases of non-obese adipose tissue was detected by semi-quantitative RT-PCR. The contents of visfatin and APN were measured by ELISA. The changes of waist circumference (WC), hip circumference HC), blood pressure (BP), fasting blood glucose (FPG) and blood lipids. Results ① There was no significant difference in adiponectin mRNA expression between obesity group and non-obese control group. The expression of APN mRNA in obesity group was significantly lower than that in non-obese control group (P <0.05) There was no significant difference in mRNA expression between the two groups of omentum and subcutaneous adipose tissue; serum visfatin obesity group was significantly higher than non-obese control group (P <0.05), APN was no significant difference between the two groups . ② The diastolic blood pressure (DBP) and SBP of obesity group were significantly higher than those of non-obese control group (P <0.01 and P <0.01), and the levels of HDL-C were significantly lower than those of non-obese control group <0.05). ③ The expression of visfatin mRNA in omental adipose tissue was positively correlated with serum visfatin, BMI and TG. APN mRNA expression was positively correlated with serum APN and negatively correlated with BMI and LDL-C. Conclusions The level of serum lipids in Uygur obese patients is elevated, while there is no significant difference in the expression of lipids mRNA in omental and subcutaneous adipose tissue. The expression of APN mRNA in omental adipose tissue is significantly decreased, and visfatin and adiponectin may be Play a role in Uighur central obesity-related metabolic disorders.
