本实验观察了A-LAK细胞的体外扩增及其抗肿瘤作用,并与LAK细胞进行了比较。结果表明,A-LAK细胞在短期内大量扩增,在培养第7天时达到增殖高峰,A-LAK细胞增加12.02倍,而LAK细胞增加2.69倍,培养3周后A-LAK细胞增加53.50倍,而LAK细胞仅增加4.45倍。18小时LDH-L释放试验表明,A-LAK细胞对Anip973和K562细胞均显示较高的杀伤活性,与LAK细胞相比有显著差异(P<0.05)。通过FACS细胞表型分析显示,A-LAK细胞大量表达CD16、CD56抗原,提示A-LAK细胞可能来源于LGL-NK细胞群。
In this experiment, the in vitro expansion of A-LAK cells and its anti-tumor effect were observed and compared with LAK cells. The results showed that A-LAK cells expanded rapidly in the short-term, reached the peak of proliferation on the 7th day of culture, A-LAK cells increased 12.02-fold, while LAK cells increased 2.69-fold, and A-LAK was cultured after 3 weeks. The cells increased 53.50-fold while LAK cells only increased 4.45-fold. The 18-hour LDH-L release assay showed that A-LAK cells showed higher killing activity against Anip973 and K562 cells, which was significantly different from that of LAK cells (P<0.05). The phenotypic analysis of FACS cells showed that A-LAK cells express a large number of CD16 and CD56 antigens, suggesting that A-LAK cells may be derived from LGL-NK cell population.
