目的探讨子宫内膜癌组织中COX-2表达的上调与肿瘤细胞增殖的关系。方法分析60例子宫内膜癌患者,应用蛋白印迹法分别测定子宫内膜癌组织中COX-2、VEGF的蛋白表达水平,应用流式细胞技术分析肿瘤细胞凋亡、增殖情况。其中无肌层浸润组中30例子宫内膜癌病例均为早期无肌层侵袭和淋巴转移者;肌层浸润组中30例子宫内膜癌病例均有深肌层侵袭和(或)伴有淋巴转移者。对照组入选病例均无子宫内膜病变。结果①研究组1和2中COX-2、VEGF蛋白表达水平均高于对照组,且有统计学意义(P<0.05);②研究组1和2之间COX-2、VEGF的蛋白表达水平,且有显著统计学意义(P<0.05);③研究组1和2中肿瘤组织PI指数均高于对照组,且有统计学意义。结论子宫内膜癌组织中COX-2的上调促进细胞增殖,抑制细胞凋亡,促进新生血管和淋巴管生成,从而影响子宫内膜癌肌层浸润和淋巴转移。
Objective To investigate the relationship between the up-regulation of COX-2 expression and the proliferation of tumor cells in endometrial carcinoma. Methods 60 cases of endometrial carcinoma were analyzed. The protein expression of COX-2 and VEGF in endometrial carcinoma was detected by Western blotting. The apoptosis and proliferation of tumor cells were analyzed by flow cytometry. Among them, 30 cases of endometrial carcinoma without myometrial infiltration group were all early myometrial invasion and lymph node metastasis; 30 cases of endometrial carcinoma with myometrial infiltration group had deep myometrial invasion and / or accompanied Lymphatic metastasis. There were no endometrial lesions in the control group. Results ① The expression levels of COX-2 and VEGF in study group 1 and 2 were significantly higher than those in control group (P <0.05); ② The protein expression of COX-2 and VEGF in study group 1 and 2 (P <0.05). ③ The PIs of tumor tissue in study groups 1 and 2 were higher than those in control group, and there was statistical significance. Conclusion Upregulation of COX-2 in endometrial carcinoma can promote cell proliferation, inhibit apoptosis, promote neovascularization and lymphangiogenesis, and thus affect myometrial invasion and lymphatic metastasis in endometrial cancer.
