该文鉴定了一种提取物具有维持和增强细胞存活与分化的能力,这将在细胞生物学研究中得到应用。制备四种卵细胞提取物,雄鼠的脾细胞被提取物渗透并培养,然后输注给雌鼠。在移植后雌鼠外周血中GFP阳性细胞和骨髓细胞中的Y染色体被检测,对比脾细胞在体内存活与分化的不同能力。移植蛋白和全蛋提取液处理的脾细胞的雌鼠外周血检测到GFP阳性细胞,在移植后120 d,雌鼠骨髓中检测到的Y染色体阳性率与外周血中的GFP阳性细胞率呈正比。用蛋白和全蛋提取液渗透脾细胞后,脾细胞在雌鼠体内存活能力强于其他提取液。结果表明,鸡蛋白提取液有维持并增强脾细胞存活的作用。因此蛋白提取物可在将来用于干细胞功能的维持。
This article identifies an extract that has the capacity to maintain and enhance cell survival and differentiation, which will be used in cell biology research. Four kinds of egg extract were prepared, and the spleen cells of the male mouse were infiltrated and cultured with the extract, and then infused into the female rats. The Y chromosome in GFP-positive cells and myeloid cells in the peripheral blood of the female rats after transplantation was tested to compare the different capacities of spleen cells to survive and differentiate in vivo. GFP-positive cells were detected in the peripheral blood of female mice spleen cells treated with the allografted protein and the whole egg extract. The positive rate of Y chromosome detected in the bone marrow of female mice at 120 d after transplantation was directly proportional to the rate of GFP-positive cells in the peripheral blood . After the spleen cells were infiltrated with the protein and the whole egg extract, the spleen cells survived in the female rats more strongly than other extracts. The results showed that the egg white extract has the effect of maintaining and enhancing the survival of spleen cells. Protein extracts can therefore be used in the future for the maintenance of stem cell function.
