目的对蛇床Cnidium monnieri干燥成熟果实中的色原酮类化学成分进行研究,并探讨其对类成骨细胞UMR106增殖的影响。方法采用多种色谱柱技术进行分离纯化,通过波谱分析鉴定化合物结构。对所分离得到的化合物进行类成骨细胞UMR106增殖活性的测试。结果从蛇床子75%乙醇提取物中分离得到10个色原酮类化合物,分别鉴定为cnidimoside A(1)、cnidimol B(2)、前胡色原酮(3)、5,7-二羟基色原酮(4)、5-O-甲基维斯阿米醇(5)、5-O-甲基维斯阿米醇苷(6)、亥茅酚(7)、2,5-二甲基-7-羟基色原酮(8)、升麻素(9)、5-羟基色原酮-7-O-β-葡萄糖苷(10)。在样品溶液浓度为0.10 nmol/L条件下,化合物1、5、9、10对类成骨细胞UMR106的增殖促进率分别是30.23%、31.56%、35.29%和33.36%。结论化合物3~10为首次从蛇床属植物中分离得到。化合物1、5、9、10能一定程度上促进UMR106细胞的增殖。
OBJECTIVE: To study the chemical constituents of chromone in dried ripe fruit of Cnidium monnieri and explore its effect on the proliferation of osteoblast-like UMR106. Methods Various chromatographic techniques were used to separate and purify the compounds. The structures of the compounds were identified by spectral analysis. The isolated compounds were tested for osteoblastic UMR106 proliferative activity. Results Ten chromone compounds were isolated from 75% ethanol extract of Fructus Cnidii and identified as cnidimoside A (1), cnidimol B (2), prochordone (3), 5,7-dihydroxy (4), 5-O-methylvariamide (5), 5-O-methylvishisainin (6), hainol (7), 2,5-bis Methyl-7-hydroxychromone (8), Cimilil (9), 5-hydroxychromophorone-7-O-β-glucoside (10). Under the concentration of 0.10 nmol / L, the proliferation of the osteoblast-like cells UMR106 with compound 1, 5, 9 and 10 were 30.23%, 31.56%, 35.29% and 33.36%, respectively. Conclusion Compounds 3 ~ 10 were isolated from Cnidium for the first time. Compounds 1, 5, 9 and 10 promoted the proliferation of UMR106 cells to a certain extent.
