目的:研究白花蛇舌草水提取物对多药耐药白血病细胞HL-60/ADR的抑制作用及机制。方法:采用MTT比色试验观察白花蛇舌草水提取物对多药耐药细胞HL-60/ADR的抑制作用。采用光镜、电镜技术观察细胞形态结构的改变,利用流式细胞术(FCM)检测细胞凋亡率。结果:①白花蛇舌草水提取物能明显抑制HL-60/ADR细胞的生长。②形态学改变,呈典型的凋亡特征。③流式细胞术证实,凋亡率也呈时间和剂量的依赖性。结论:白花蛇舌草水提取物对多药耐药细胞HL-60/ADR的生长具有极强的抑制作用,诱导其凋亡是其主要机制。
Objective: To study the inhibitory effect and mechanism of aqueous extract of Hedyotic diffusa on HL-60 / ADR multidrug resistance leukemia cells. Methods: MTT colorimetric assay was used to observe the inhibitory effect of extract of Hedyotic diffusa on HL-60 / ADR multidrug resistance cells. The morphological changes of cells were observed by light microscope and electron microscope. The apoptosis rate was detected by flow cytometry (FCM). Results: ① Hedyotis diffusa extract could significantly inhibit the growth of HL-60 / ADR cells. ② morphological changes, typical apoptotic characteristics. Flow cytometry confirmed that the apoptosis rate was also time-and dose-dependent. Conclusion: The extract of Hedyotic diffusa has a strong inhibitory effect on the growth of multidrug-resistant cells HL-60 / ADR, and inducing apoptosis is the main mechanism.
