目的研究蝎毒多肽对胃癌SGC-7901细胞侵袭转移的作用。方法设立空白组和5个不同质量浓度(蝎毒多肽5,10,20,40,80μg·m L~(-1))实验组。以噻唑蓝法检测蝎毒多肽对细胞活性和增殖的影响,以划痕实验和Transwell实验观察其对细胞侵袭及转移能力的影响,以免疫印迹法检测其对基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)的影响。结果与空白组相比,蝎毒多肽可以明显抑制胃癌SGC-7901细胞的增殖,且呈现时间-浓度依赖性(P<0.05)。2个不同质量浓度(10,20μg·m L~(-1))蝎毒多肽侵袭细胞的数量分别为(35.8±5.53),(19.3±2.13),与空白组(48.3±7.34)比较差异有统计学意义(P<0.05),表明其可抑制细胞的侵袭能力。2个浓度实验组的迁移抑制率分别为(14.7±1.23)%,(28.5±3.35)%,实验组较空白组明显降低(P<0.05)。2个不同质量浓度(10,20μg·L~(-1))蝎毒多肽作用于胃癌SGC-7901细胞12 h后,与空白组比较均可抑制细胞MMP-9和MMP-2的表达。结论蝎毒多肽通过抑制MMP-2及MMP-9的表达从而抑制胃癌SGC-7901细胞的侵袭转移。
Objective To study the effect of scorpion venom peptide on invasion and metastasis of gastric cancer cell line SGC-7901. Methods A blank group and 5 experimental groups with different concentrations of 5, 10, 20, 40, 80 μg · m L -1 were screened. The effect of scorpion venom peptide on cell viability and proliferation was detected by thiazolyl blue assay. The effect of scorpion venom peptide on cell invasion and metastasis was observed by scratch test and Transwell assay. The effect of scorpion venom polypeptide on the activity of matrix metalloproteinase-2 ) And matrix metalloproteinase-9 (MMP-9). Results Compared with the blank group, scorpion venom peptide could significantly inhibit the proliferation of gastric cancer SGC-7901 cells in a time-concentration-dependent manner (P <0.05). The number of invasive cells of two scorpion venom peptides with different concentrations (10, 20μg · m L -1) were (35.8 ± 5.53) and (19.3 ± 2.13), respectively, which were significantly different from those in the blank group (48.3 ± 7.34) Statistical significance (P <0.05), indicating that it can inhibit the invasion of cells. Migration inhibition rate of the two concentration experimental groups were (14.7 ± 1.23)% and (28.5 ± 3.35)%, respectively, and the experimental group was significantly lower than the blank group (P <0.05). Two different concentrations of scorpion venom peptide (10,20μg · L -1) could inhibit the expression of MMP-9 and MMP-2 in gastric cancer SGC-7901 cells for 12 h compared with the blank group. Conclusion Scorpion venom polypeptide can inhibit the invasion and metastasis of gastric cancer cell line SGC-7901 by inhibiting the expression of MMP-2 and MMP-9.
