当前用于确定生物大分子结构的基本的可靠的方法是X射线晶体衍射,但它并不能肯定大分子的全部结构。例如对胰岛素B链的第4个残基谷氨酰胺和第22个残基精氨酸国外报道都有取两种构象的可能。我国X射线晶体衍射的结果认为B22确有这种可能性,而B4的第二种构象的电子密度很可能代表水分子。我们采用Levitt的经验势对此问题进行了探讨。
The basic reliable method currently used to determine the structure of a biological macromolecule is the X-ray crystal diffraction, but it does not confirm the overall structure of the macromolecule. For example, the second residue of glutamine in the B chain of insulin and the 22th residue of arginine have been reported in both foreign countries. As a result of X-ray crystal diffraction in our country, B22 does have the possibility that the electron density of the second conformation of B4 probably represents water molecules. We use Levitt’s experience to explore this issue.
