目的建立iASPP基因的实时荧光定量检测方法,并观察原发性肝癌(hepatocellular carcinoma,HCC)组织中iASPP mRNA的表达情况,分析临床病理学因素在肝癌iASPP基因和扩增中的作用。方法对46例病理学证实的HCC患者,利用荧光定量RT-PCR检测肝癌组织及癌旁组织iASPP mRNA表达,比较不同临床病理学类型肝癌中该基因表达差异。结果 46例肝癌组织中有33例的iASPP表达水平高于癌旁组织,占71.4%,表达差异有统计学意义[癌组织(16.00±10.31),癌旁组织(0.63±0.19),P<0.01);在不同肿瘤大小、病理分级中有统计学差异(P<0.05)],在患者的性别、年龄、癌栓形成、淋巴结转移分组中差异无统计学意义(P>0.05)。结论肝癌组织中存在iASPP基因的高表达;iASPP基因表达与肿瘤大小、病理分级有关。
Objective To establish real-time fluorescence quantitative detection of iASPP gene and to observe the expression of iASPP mRNA in primary hepatocellular carcinoma (HCC) tissues and to analyze the role of clinicopathological factors in iASPP gene amplification. Methods The expression of iASPP mRNA in hepatocellular carcinoma and adjacent non-cancerous tissues was detected by fluorescence quantitative RT-PCR in 46 HCC patients with pathologically confirmed pathological changes, and the differences in gene expression between different clinical pathological types of HCC were compared. Results The expression of iASPP in 33 of 46 HCC tissues was significantly higher than that in paracancer tissues, accounting for 71.4% (16.00 ± 10.31), paracancer tissues (0.63 ± 0.19), P <0.01 ). There was a statistically significant difference in tumor size and pathological grade (P <0.05). There was no significant difference in gender, age, tumor thrombus formation and lymph node metastasis between the two groups (P> 0.05). Conclusion The high expression of iASPP gene exists in HCC tissues. The expression of iASPP gene is related to tumor size and pathological grade.
