目的和方法:利用人手术切除胃标本,应用Pronase-EDTA法进行人正常胃粘膜细胞的分离及孵育,并采用羟自由基建立细胞损伤模型,观察八肽生长抑素(SS-8)对细胞损伤的影响。结果:(1)采用Pronase-EDTA法分离人胃粘膜上皮细胞,每4cm×5cm大小胃粘膜可分离细胞数为24×106个,细胞存活率达90%以上。(2)Fe2+-H2O2体系产生的羟自由基(OH·)可直接导致人胃粘膜细胞存活率下降,细胞乳酸脱氢酶漏出量及细胞丙二醛含量增加,以SS-8预处理可减轻上述改变。结论:SS-8可减轻OH·对孵育状态下人胃粘膜细胞的损伤,作用与其减轻细胞的脂质过氧化程度有关。
OBJECTIVE: To investigate the effect of SS-8 on the proliferation of human gastric mucosa cells and to explore the possible mechanism of gastric mucosal injury.Methods: The impact of injury. Results: (1) Human gastric mucosal epithelial cells were isolated by Pronase-EDTA method. The number of detachable cells per 4cm × 5cm gastric mucosa was 24 × 106, and the cell survival rate was over 90%. (2) Hydroxyl radical (OH ·) produced by Fe2 + -H2O2 system can directly lead to the decrease of cell viability of human gastric mucosa, cell leakage of lactate dehydrogenase and increase of malondialdehyde (MDA) The above change. Conclusion: SS-8 can reduce the OH · on human gastric mucosal injury under incubation, and its role in reducing cell lipid peroxidation level.
