目的:研究益肺汤对人肺腺癌耐顺铂细胞A549/DDP裸鼠移植瘤多药耐药逆转作用的机理。方法:建立A549/DDP裸鼠移植瘤模型,分为6组,每组10只,分别以生理盐水(对照组)、益肺汤低、中、高剂量(益肺汤低、中、高剂量组)、益肺汤联合顺铂(联合组)、顺铂(顺铂组)进行干预,观察移植瘤的生长状况,隔天测量一次瘤块体积,给药21天后,处死小鼠取瘤组织,称重,实时荧光定量PCR检测多药耐药蛋白(MRP)基因的表达。结果:对照组的移植瘤体积最大,增长最快,与其他组比较,差异均有统计学意义(P<0.05)。联合组与其他药物组比较,移植瘤体积最小,抑瘤率最高,差异均有统计学意义(P<0.05)。益肺汤低剂量组与中、高剂量组比较,差异均有统计学意义(P<0.05),而后二者比较,差异无统计学意义(P>0.05)。益肺汤低剂量组与顺铂组比较,差异无统计学意义(P>0.05)。q=1.37,益肺汤与顺铂在抑制肿瘤生长上有协同作用。实时荧光定量PCR检测的结果显示,益肺汤低、中、高剂量组及联合组的MRP基因的表达量均低于对照组,而顺铂组MRP基因表达量却高于对照组,差异均有统计学意义(P<0.05)。联合组、中剂量组及高剂量组三者相互比较,差异均无统计学意义(P>0.05),三者与益肺汤低剂量组比较,差异均有统计学意义(P<0.05)。结论:益肺汤能逆转A549/DDP移植瘤的多药耐药性,其逆转机制可能是通过下调MRP基因的表达来实现的。
Objective: To study the mechanism of Yifei Decoction reversing the multidrug resistance in human lung adenocarcinoma A549 / DDP xenografts. Methods: The A549 / DDP xenografted model of nude mice was established and divided into 6 groups with 10 rats in each group. The rats in each group were treated with saline (control group), Yifei decoction low, middle and high dose Group), Yifei Decoction combined with cisplatin (combination group) and cisplatin (cisplatin group) were given interventions to observe the growth of the tumor. The size of the tumor mass was measured every other day. Yifei Decoction low-dose group and the medium-high dose group, the difference was statistically significant (P <0.05), and then the two compared, the difference was not statistically significant (P> 0.05). Yifu Tang low-dose group and cisplatin group, the difference was not statistically significant (P> 0.05). q = 1.37, Yifei Decoction and cisplatin in the inhibition of tumor growth have a synergistic effect. The results of real-time quantitative PCR showed that MRP gene expression of Yifei Decoction low, middle and high dose group and combination group were lower than that of control group, while the expression of MRP gene in cisplatin group was higher than that of control group There was statistical significance (P <0.05). There was no significant difference among the three groups (P> 0.05). The difference between the three groups was statistically significant (P <0.05). Conclusion: Yifei Decoction can reverse the multidrug resistance of A549 / DDP xenografts, and its reversal mechanism may be through the down-regulation of MRP gene expression.
