目的:探讨同型半胱氨酸(HCY)对大鼠主动脉内皮细胞(EC)脂质过氧化的影响。方法:取体重150~200g健康雄性Wistar大鼠胸主动脉,以组织贴块法进行EC培养,第5代用于实验。各实验组及对照组均为 6孔细胞,实验组加入 5mmol/L HCY或 5 mmol/L HCY+ 25μmol/L Cu~2,对照组不加 HCY。分别在不同时限观察细胞形态并测定丙二醛(MDA)浓度,超氧化物歧化酶(SOD)活性和乳酸脱氢酶(LDH)浓度。结果:HCY可引起EC形态变化,使培养液中LDH及MDA浓度增加,而SOD活性下降,MDA浓度与SOD活性呈负相关(r=-0.838,P< 0. 05),在有 Cu~2+存在时,上述改变更为明显。结论: HCY引起EC脂质过氧化可能是导致动脉粥样硬化的原因之一。
Objective: To investigate the effect of homocysteine (HCY) on lipid peroxidation in rat aortic endothelial cells (ECs). Methods: The thoracic aorta of healthy male Wistar rats weighing 150 ~ 200g were taken and cultured by tissue patch method. The fifth passage was used for the experiment. The experimental group and the control group were 6-hole cells, the experimental group by adding 5mmol / L HCY or 5 mmol / L HCY + 25μmol / L Cu ~ 2, the control group without HCY. The morphological changes of cells were observed at different time points and the concentrations of malondialdehyde (MDA), superoxide dismutase (SOD) and lactate dehydrogenase (LDH) were measured. Results: HCY could induce morphological changes of EC, increase the concentration of LDH and MDA, and decrease the activity of SOD in culture medium. The concentration of MDA was negatively correlated with the activity of SOD (r = -0.838, P <0.05) ~ 2 + exists, the above change is more obvious. Conclusions: HCY induced lipid peroxidation of EC may be one of the causes of atherosclerosis.
