目的:比较研究芦丁、槲皮素、槐米黄酮部位和槐米药材提取物对高糖诱导的内皮细胞损伤的修复作用。方法:采用含有33mmol·L-1葡萄糖和1%FBS的DMEM低糖培养基处理人脐静脉HUVEC内皮细胞,建立细胞损伤模型,采用NO试剂盒检测各样品对模型内皮细胞NO分泌量的影响;采用MTT法测定细胞活力。结果:芦丁、槲皮素、槐米黄酮部位和槐米药材提取物均可不同程度地升高模型HUVEC内皮细胞NO分泌量,不同程度地增强细胞活力。结论:芦丁和槲皮素可能为槐米修复内皮细胞损伤的活性成分。
OBJECTIVE: To compare the effects of rutin, quercetin, sophoridine and extract of Sophora japonica L. on the injury of endothelial cells induced by high glucose. Methods: Human umbilical vein endothelial cells (HUVECs) were treated with DMEM low glucose medium containing 33mmol·L-1 glucose and 1% FBS to establish the cell injury model. The NO kit was used to detect the effect of each sample on NO secretion in the model endothelial cells. MTT assay of cell viability. Results: Rutin, quercetin, sophoricin and Scutellaria barbata extract could increase NO secretion of HUVEC endothelial cells to varying degrees and enhance cell viability in different degree. CONCLUSION: Rutin and quercetin may be the active components of Huai Mi for repairing endothelial cell injury.
