目的检测大鼠心、肝、脾、肾β肌动蛋白(β-actin)mRNA在死后不同时间的表达,为死亡时间的推断寻求客观依据。方法大鼠麻醉致死后置于18℃温度控制系统模拟死后改变,在不同时间点抽提相应组织总mRNA,利用RT-PCR技术检测β-actin mRNA的表达水平变化并对扩增产物进行半定量分析。结果大鼠心脏β-actin mRNA在死后10天内都可以检测到,肝脏β-actin mRNA在死后4天内都可以检测到,脾脏β-actin mRNA在死后12天内都可以检测到,肾脏β-actin mRNA在死后8天内都可以检测到。结论观测死后心、肝、脾、肾β-actin mRNA的变化,可为死亡时间推断提供客观依据。
Objective To detect the expression of β-actin mRNA in rat heart, liver, spleen and kidney at different time after death, and to find an objective basis for the estimation of death time. Methods The rats were killed by anesthesia and then placed in a temperature control system of 18 ℃. The total mRNA was extracted from the corresponding tissues at different time points. The expression level of β-actin mRNA was detected by RT-PCR and half Quantitative analysis. Results β-actin mRNA in rat heart can be detected within 10 days after death, β-actin mRNA in liver can be detected within 4 days after death, β-actin mRNA in spleen can be detected within 12 days after death, The -actin mRNA is detectable within 8 days of death. Conclusion The change of β-actin mRNA in the heart, liver, spleen and kidney after death can provide an objective basis for the estimation of death time.
