目的研究全反式维甲酸(ATRA)衍生物[N-(3-三氟甲基苯基)维甲酰胺,NTFMP]对人肺腺癌A549细胞株迁移能力的影响及其可能机制。方法应用细胞迁移实验分析NTFMP对A549细胞迁移能力的影响,Western blot检测NTFMP对肌球蛋白轻链激酶(MLCK)的表达和肌球蛋白轻链(MLC)磷酸化的影响。结果 1、7.5 mg/L NTFMP均可抑制A549细胞的迁移,迁移距离随药物浓度的增加而明显下降,7.5 mg/L NTFMP显著抑制MLCK的表达和MLC磷酸化,而对MLC蛋白表达无明显作用。运用双变量相关分析,发现A549细胞的迁移与细胞内MLCK含量和MLC磷酸化程度呈正相关。结论 NTFMP对A549细胞迁移有抑制作用,其机制可能与MLCK表达和MLC磷酸化下调有关。
Objective To investigate the effect of all-trans retinoic acid (ATRA) derivative [N- (3-trifluoromethylphenyl) retinamide and NTFMP] on the migration of human lung adenocarcinoma A549 cell line and its possible mechanism. Methods The effect of NTFMP on the migration of A549 cells was analyzed by cell migration assay. The effect of NTFMP on the expression of myosin light chain kinase (MLCK) and phosphorylation of myosin light chain (MLC) was detected by Western blot. Results 1, 7.5 mg / L NTFMP could inhibit the migration of A549 cells. The migration distance decreased obviously with the increase of drug concentration. 7.5 mg / L NTFMP significantly inhibited MLCK expression and MLC phosphorylation, but had no effect on MLC protein expression . Using bivariate correlation analysis, it was found that A549 cell migration and intracellular MLCK content and MLC phosphorylation was positively correlated. Conclusion NTFMP can inhibit the migration of A549 cells. The mechanism may be related to the down-regulation of MLCK expression and MLC phosphorylation.
