大孔吸附树脂纯化倒卵叶五加中紫丁香苷的工艺优选

目的:优选大孔吸附树脂纯化倒卵叶五加中紫丁香苷的工艺条件。方法:采用HPLC测定紫丁香苷含量,通过静态吸附-洗脱试验考察大孔树脂型号,利用单因素试验考察上样量、吸附流速、洗脱机浓度及用量对纯化工艺的影响。结果:选用XDA-1型大孔树脂,其纯化工艺条件为上样量每g树脂吸附3.5 mg紫丁香苷,吸附流速1 BV.h-1,用9 BV 40%乙醇于2BV.h-1流速洗脱,紫丁香苷纯度由5.4%提高到23.1%。结论:XDA-1型大孔树脂可用于纯化倒卵叶五加提取液,紫丁香苷收率和纯度均达满意效果。 OBJECTIVE: To optimize the process condition of purification of syringylidene glucoside in Oviductus polyporus by macroporous adsorption resin. Methods: The content of syringin was determined by HPLC. The macroporous resin model was investigated by static adsorption - elution test. The effect of sample volume, adsorption flow rate, concentration of eluate and dosage on the purification process was investigated by single factor test. Results: Using XDA-1 macroporous resin, the purification conditions were as follows: sample loading was 3.5 mg syringin per gram of resin, adsorption flow rate was 1 BV.h-1, and 9 BV 40% ethanol was used in 2BV.h-1 Flow elution, syringin purity increased from 5.4% to 23.1%. Conclusion: XDA-1 macroporous resin can be used to purify the extract of Populus obscurus. The yield and purity of syringin reach satisfactory results.
