目的检测梅毒血清固定患者Th17细胞免疫调节网络相关84种基因的表达水平,初步揭示Th17细胞在梅毒血清固定中的作用机理。方法收集2015年9—11月深圳市慢性病防治中心的8例梅毒血清固定患者(固定组)及8例健康人(健康对照组)的外周血,提取总RNA,逆转录合成c DNA,应用实时定量聚合酶链反应Array法(PCR-Array)检测Th17细胞免疫调节网络相关84种基因m RNA的表达水平,以表达差异(上调或下调)大于2倍的基因作为有意义的差异表达基因。结果固定组与健康对照组相比表达上调的基因为Foxp3和IL-10;表达下调基因13种,分别为:CCL22、CSF2、CSF3、CXCL6、IL-17A、IL-17D、IL-21、IL-23R、IL-9、IRF4、RORA、RORC、STAT3。结论 Th17细胞免疫调节网络中基因的异常表达可能与梅毒血清固定相关。
Objective To detect the expression levels of 84 genes related to Th17 cell immune regulation network in patients with syphilis and to reveal the mechanism of Th17 cells in syphilis serum fixation. METHODS: Peripheral blood samples of 8 patients with syphilis (fixed group) and 8 healthy individuals (healthy control group) from Shenzhen Center for Chronic Disease Prevention and Control from September to November in 2015 were collected. Total RNA was extracted and cDNA was reverse transcribed into cDNA. Quantitative polymerase chain reaction (PCR-Array) was used to detect the mRNA expression level of 84 genes related to Th17 cell immune regulatory network, and the genes with more than 2-fold difference (up-regulated or down-regulated) were used as significant differentially expressed genes. Results The expression of Foxp3 and IL-10 were up-regulated in the fixed group compared with the healthy control group, while the expression of 13 genes was down-regulated in the fixed group, which were CCL22, CSF2, CSF3, CXCL6, IL-17A, IL-17D, IL- -23R, IL-9, IRF4, RORA, RORC, STAT3. Conclusion Abnormal expression of Th17 cell immunoregulatory network may be related to syphilis serum fixation.
