目的利用高效液相色谱(HPLC)建立羟基柠檬酸(hydroxycitric acid,HCA)功效成分的分析方法,并建立功能食品中非法添加利莫那班(rimonabant)和西布曲明(sibutramine)的同时检测方法。方法样品经过超声提取,分别以99%0.1%磷酸水溶液?1%甲醇为流动相,在210nm检测波长下进行HCA检测;以100%甲醇为流动相,在223nm波长下进行利莫那班和西布曲明同时检测。结果 HCA在0.001~1.0mg/mL范围内线性关系良好,平均添加回收率为90.49%~103.90%。利莫那班和西布曲明在0.01~20??g/mL范围内线性关系良好,检出限分别达到1ng/mL和3ng/mL。结论该方法简单、分析速度快、准确度高,能够定量检测功能食品中羟基柠檬酸含量,以及非法添加利莫那班和西布曲明,实际用于该类功能食品的质量监控效果良好。
OBJECTIVE To establish a method for the determination of the active ingredient of hydroxycitric acid (HCA) by high performance liquid chromatography (HPLC) and to establish the simultaneous detection of illicit addition of rimonabant and sibutramine in functional foods method. Methods Samples were sonicated and extracted with 99% 0.1% aqueous solution of phosphoric acid and 1% methanol respectively. HCA assay was carried out at the detection wavelength of 210 nm. With 100% methanol as the mobile phase, Buqu Ming simultaneous detection. Results The linearity of HCA in the range of 0.001-1.0 mg / mL was good with the average recovery of 90.49% -103.90%. The linear relationship between rimonabant and sibutramine in the range of 0.01 ~ 20 μg / mL was good with detection limits of 1 ng / mL and 3 ng / mL, respectively. Conclusions This method is simple, rapid and accurate. It can quantitatively detect hydroxycitric acid content in functional foods and ilimonabine and sibutramine illegally. The quality control of such functional foods is actually good.
