目的: 研究人脐静脉内皮细胞(HUVEC)感染汉坦病毒 (Hantavirus, HTV)后应激反应的规律及意义. 方法: 采用免疫细胞化学染色法和核酸分子原位杂交, 检测热休克蛋白 70(HSP70)的表达; 用RT PCR观察HSP70mRNA水平变化的规律. 结果:HTV感染HUVEC后, 免疫细胞化学染色法可检出HSP70呈高表达, 原位杂交发现细胞质内有HSP70mRNA的阳性信号. 感染后不同时间点RT PCR的结果表明, 与对照组相比较, HSP70mRNA的水平在感染后迅速升高并持续高表达(P<0. 05). 结论:HTV可诱导HUVEC高表达HSP70. HSP70可能具有抑制病毒复制和保护内皮细胞的作用.
Objective: To study the regularity and significance of stress response after Hantavirus (HUV) infection in human umbilical vein endothelial cells (HUVEC) .Methods: Immunocytochemistry and in situ hybridization were used to detect the expression of heat shock protein 70 HSP70) were detected by RT-PCR.Results: The expression of HSP70 was detected by immunocytochemical staining after HTV infection in HUVEC.HSP70 mRNA was found in the cytoplasm by in situ hybridization.After infection, The result of RT-PCR showed that the level of HSP70 mRNA increased rapidly and persistently high after infection (P <0.05) .Conclusion: HTV can induce high expression of HSP70 in HUVEC.HSP70 may have inhibitory effect on virus The role of endothelial cells in replication and protection.
