鬼臼经乙醚脱脂后用热水抽提 ,用蛋白酶法和 Sevag法相结合除去蛋白质 ,乙醇分级沉淀 ,经DEAE- Sepharose和 Superdex G- 75或 Sephacryl S- 2 0 0 HR柱层析纯化得到两种多糖组分 EPS- A和EPS- B。经分析该两种多糖均为单一组分。用分子筛层析法测定 EPS- A和 EPS- B的分子量分别为 15 k D和 175 k D。纸层析和气相层析分析得知 ,EPS- B含有木糖、阿拉伯糖、鼠李糖、葡萄糖、甘露糖和半乳糖 ,其摩尔比为 0 .37∶ 2 .2 0∶ 0 .5 7∶ 9.83∶ 1.0∶ 3.42 ,而 EPS- A中仅含葡萄糖。体内试验结果表明 ,EPS-B多糖对小鼠腹水肝癌细胞生长有一定的抑制作用
Podophyllum was defatted with diethyl ether and extracted with hot water. Proteins were combined with Sevag method to remove proteins. Ethanol was fractionated and precipitated, and purified by DEAE-Sepharose and Superdex G-75 or Sephacryl S-200 HR column chromatography to obtain two kinds of proteins. Polysaccharide components EPS-A and EPS-B. Both polysaccharides were analyzed as single components. The molecular weights of EPS-A and EPS-B were determined by molecular sieve chromatography to be 15 kD and 175 kD, respectively. Paper chromatography and gas chromatography analysis showed that EPS-B contained xylose, arabinose, rhamnose, glucose, mannose, and galactose in a molar ratio of 0.37:2.20:0.57. : 9.83: 1.0: 3.42, while EPS-A contains only glucose. In vivo test results showed that EPS-B polysaccharides inhibited the growth of mouse ascites hepatoma cells.
