目的建立玉簪花总皂苷的含量测定方法。方法采用可见分光光度法,以香草醛-冰醋酸-浓硫酸为显色剂,支脱皂苷元为对照品,在450 nm波长下测定吸光度,标准曲线法计算供试品中总皂苷的含量。结果支脱皂苷元的质量浓度在14~70 mg/L范围内与吸光度呈良好的线性关系(r=0.999 2),平均加样回收率为96.8%,RSD=2.97%(n=6)。结论该法操作简便、准确、灵敏、重复性好,可用于玉簪花药材中总皂苷的含量测定。
Objective To establish a method for determining the content of total saponins in flowers of hosta. Methods Visible spectrophotometric method was used. Vanillin-glacial acetic acid-concentrated sulfuric acid was used as the color reagent. The absorbance was measured at the wavelength of 450 nm. The standard curve method was used to calculate the total saponin content of the test sample. Results The mass concentration of sapogenin was linearly correlated with the absorbance in the range of 14-70 mg/L (r=0.999 2). The average recovery was 96.8% and the RSD was 2.97% (n=6). Conclusion The method is simple, accurate, sensitive and reproducible. It can be used for the determination of total saponins in Radix Astragali.
