目的测定盐酸小檗碱对小鼠黑色素瘤B16细胞的诱导分化作用。方法以MTT法测定盐酸小檗碱对B16细胞增殖的抑制作用,通过克隆形成实验、细胞黑色素含量的测定以及B16细胞体内成瘤能力的测定观察盐酸小檗碱对B16细胞的诱导分化作用。结果盐酸小檗碱对B16细胞具有明显的增殖抑制作用;并使B16细胞生长缓慢,平铺不重叠,呈正常上皮样细胞分化,细胞的克隆形成能力降低,细胞内的黑色素生成能力增强,C57/BL小鼠体内成瘤能力降低。结论盐酸小檗碱对B16细胞具有诱导分化作用。
Objective To determine the effect of berberine on the differentiation of mouse melanoma B16 cells. Methods The inhibitory effect of berberine hydrochloride on the proliferation of B16 cells was determined by MTT assay. The effect of berberine hydrochloride on differentiation and differentiation of B16 cells was observed by colony formation assay, determination of melanin content and tumorigenicity of B16 cells in vivo. Results Berberine hydrochloride had a significant inhibitory effect on B16 cells proliferation. B16 cells grew slowly and did not overlap, showing normal epithelioid cell differentiation, decreased cell clonogenic capacity and enhanced intracellular melanin production. C57 / BL mice reduced tumorigenicity in vivo. Conclusion Berberine can induce the differentiation of B16 cells.
