目的探讨金雀异黄素(GEN)和树突状细胞瘤苗(DC疫苗)对荷瘤小鼠H22癌细胞的免疫抑制作用。方法采用共聚焦显微镜、流式细胞技术检测H22细胞凋亡的情况,蛋白免疫印迹(Western-blot)检测凋亡相关基因Bax、caspase-3、bcl-2蛋白的表达。结果共聚焦显微镜观察到GEN DC疫苗诱导凋亡的H22细胞;与1640对照组凋亡率(3.019±0.327)比较,GEN DC组总凋亡率(14.488±1.660)明显增高;GEN DC疫苗诱导可上调H22细胞bax、caspase-3基因的表达而下调bcl-2基因表达。结论金雀异黄素和DC疫苗对肝癌细胞的生长有一定抑制作用。
Objective To investigate the immunosuppressive effects of genistein (GEN) and dendritic cell tumor vaccine (DC vaccine) on tumor-bearing mice H22 cancer cells. Methods The apoptosis of H22 cells was detected by confocal microscopy and flow cytometry. The protein expressions of Bax, caspase-3 and bcl-2 were detected by Western-blot. Results The apoptotic rate of H22 cells induced by GEN DC vaccine was observed by confocal microscopy. Compared with the control group, the total apoptosis rate of GEN-treated DCs (14.488 ± 1.660) was significantly higher than that of 1640 control (3.019 ± 0.327) Up-regulate the expression of bax and caspase-3 genes in H22 cells and down-regulate the expression of bcl-2 gene. Conclusions Genistein and DC vaccine can inhibit the growth of hepatoma cells.
